Link to “A closer look at separations” We recently bought purified proteins that I will use to build a molecular weight ladder for membrane separations. I initially dissolved these proteins in salt solution and spun in an ultracentrifuge to remove aggregations. I then sized the proteins using our Malvern Zetasizer, which gives hydrodynamic radius, assuming…
SET-UP Syringe pump: 5 mL of 7.6-nmol/100µL, 5.6 µL/min pump rate. Single Channel dialyser, 10 mm long, 0.5 mm wide, 0.3 mm high in stirred beaker of DI water Fraction Collector programed to rotate every 30 min. Set up in refrigerator, 40˚C. Figure 1: Long term dialysis clearance set up in refrigerator. Syringe pump, stirred…
Introduction The µSiM BBB model is being developed at UR with the goal to understand the impact of sepsis on the brain. Here, we use an EECM-BMEC-like cell (iSPC-derived brain endothelial cells) [1] monoculture model and an EECM-BMEC-like cell and brain pericyte-like cell (BPLC; iSPC-derived brain pericytes) [2] coculture model to optimize the concentration of…
Here are the results for air permeability testing of W501. The sample closest to the origin had one very small (very difficult to see on Brian’s scope unless viewing at 100x) pinhole, the middle sample had 3 very small pinholes. The completely intact sample had a permeability of .773 cc/(cm^2*atm*s). It seems that the pinholes…
This is a repeat of the previous array experiment, but with a higher NaHCO3 concentration (1.85 mg/mL). Also, since these experiments require a lot of samples, I decided to cut chips off of wafer skeletons. This one was w622. As before, these were not RTP’ed and they were placed in the oven at 37C. I…
For everyone conducting biological studies, this paper highlights how to think about sample size and replicates. It suggests using “SuperPlots”, which color codes data points from different experiments to better visualize the type of variability seen (i.e. random variability, experimental variability, etc…). SuperPlots: Communicating reproducibility and variability in cell biology Jim and I recommend everyone…
