Diffusion Gel Reruns

The movable cutoff gels were more or less successful, but were too dirty for publication (see knowledge page). I’ve been trying to rerun them so that they look nicer, but the darker stain that I used has brought out a few new observations. The first gel following this paragraph is the new gel. The second is the old gel for comparison. The R’s and F’s are in the same order, although I did add a new start column for ease of publishing.

The box on the first gels shows a little more clearly that the samples in the retentate are getting lighter. The circle shows that we are getting some passage of myosin in this particular gel. You can sort of make it out on the second gel too, but it is very light. Is this passage of myosin a deal breaker?? It doesn’t happen in the second experiment (R2/F2).

The rerun is cleaner, but I’m not sure why we’re seeing that high mw band (myosin, red circle). My guess is this is artifact in the gel because its not a clear band like the others and we don’t see anything there on the first one. However, the stain on this gel is darker and maybe there is a hint of myosin passing through. While myosin is big, it’s also very linear.
