TECAN absorbance for 96-well plates is topping out even with pure buffer
EDIT: This problem was solved thanks to Tejas. It turns out that Falcon BD transparent 96 well plates absorb in the UV range, and are best used for bradford assays or anything that measures absorbance higher than the UV range. Apparently they sell UV-inactive 96 well plates, but they are very expensive. Josh Winans and I jury-rigged a way to use the TECAN to measure protein at 280 using cuvettes, but that is another post, located here (INSERT POST HERE).
The following is an attempt I made at a standard curve using the Barcikowski peptide at concentrations of 20uM, 15uM, 10uM, and 0uM. I used a brand new Falcon BD transparent 96-well plate and filled each well with a total of 100uL of sample.
I’m not the only one to get this kind of signal lately from the TECAN. Has anyone ever seen anything like this? This just started happening a week ago or so.
